ABSTRACT

<b>Objective::To study on the plasma lipidomics characteristics of coronary heart disease (CHD) patients with syndrome of of intermingling of phlegm and static blood, and to find differential lipid metabolites between them and healthy volunteers. <b>Method::The plasma samples from CHD patients with syndrome of intermingling of phlegm and static blood and healthy volunteers of the same age were collected. The plasma lipidomics was carried out by UPLC-Q/TOF-MS. The plasma samples were detected under positive and negative ion modes, and the primary and secondary mass spectrometry datas were collected simultaneously, and the <italic>m</italic>/<italic>z</italic> ranges were 100-2 000 and 50-2 000, respectively. The lipidomics model was established by orthogonal partial least squares discriminant analysis (OPLS-DA). Differential lipid metabolites were identified based on multivariate statistics. <b>Result::OPLS-DA model could obviously distinguish CHD patients with syndrome of intermingling of phlegm and static blood and healthy volunteers. A total of 15 plasma differential lipid metabolites were identified, such as C16 sphinganine, phytosphingosine, <italic>N</italic>, <italic>N</italic>-dimethyl-safingol, 2-hydroxyphytanic acid, orotinichalcone, PC[18∶2(2<italic>E</italic>, 4<italic>E</italic>)/0∶0], PC(0∶0/16∶0), epitestosterone sulfate, etiocholanolone sulfate, PS[22∶1(11<italic>Z</italic>)/0∶0], PC[16∶0/20∶4(5<italic>E</italic>, 8<italic>E</italic>, 11<italic>E</italic>, 14<italic>E</italic>)], PC[19∶1(9<italic>Z</italic>)/17∶2(9<italic>Z</italic>, 12<italic>Z</italic>)], PC(16∶0/0∶0), PC(18∶0/0∶0), PS[15∶1(9<italic>Z</italic>)/22∶1(11<italic>Z</italic>)]. <b>Conclusion::There are significant differences in plasma lipid characteristics between CHD patients with syndrome of intermingling of phlegm and static blood and healthy volunteers. The plasma differential lipid metabolites are helpful for the accurate differentiation of CHD patients with syndrome of intermingling of phlegm and static blood.

ABSTRACT

Lipidomics is a new branch discipline of metabolism. The hydrophilic interaction chromatography-mass spectrometry (HILIC-MS) technology is the new study method of lipidomics. The application, operation points, special advantages, and development potential for HILIC-MS were clarified from the angles of biological sample processing method, chromatography, mass spectrometry conditions establishment, and full analysis on quality control. The feasibility of using HILIC-MS technology to solve some lipid metabolism diseases problem in clinical and scientific research of TCM has been investigated.

The recent progresses on chemical components and pharmacological activities of the genus Valerianawere summarized.Besides-essential oil, the chemical composition of Valerianais mainly focused on monoterpenoids,sesquiterpenoids,lignans, flavonoids, alkaloids, etc. Iridoids are the main chemical components ofmonoterpenoids. There are two types ofiridoidson the basis of the cyclopentane open or not. The Valerianahas been drawmuch attention for their significant sedation,spasmolysis,antidepression,antitumor, against adenosine A1 receptors and cytotoxicityactivity,and had certain function for cardiovascular disease treatment. Given to the fact of the lack of systematic review and summary of studies on the Valeriana, we summarized and analyze the study literatures on the pharmacological activity of Valerianain recent years, and providedsome basisfor further study.

ABSTRACT

Objective: To study the pharmacokinetics of salvianolic acid B (Sal B) monomer in normal and hyperlipidemic rats. Methods: The hyperlipidemic rat model was replicated successfully by using high fat diet. The detection method for microdialysis samples was established by using LC-MS. The pharmacokinetic study on Sal B in normal and hyperlipidemic rats was carried out after ig administration with low-, medium-, and high-dose (25, 50, and 100 mg/kg) Sal B using microdialysis sampling technology. The microdialysis samples were corrected with in vivo recoveries and followed by LC-MS detection. The plasma drug concentration-time curves were established by the sampling time midpoint as timeline. The pharmacokinetic fitting parameters were obtained by using non-compartment model. Results: The LC-MS detection method was sensitive, specific, and able to meet the analytical requirements. The main pharmacokinetics parameters of normal rat after ig administration with low-, medium-, and high-dose Sal B were as follows: Cmax were (38.551 ± 6.692), (95.550 ± 12.544), and (204.251 ± 20.342) ng/mL, respectively, tmax were (1.125 ± 0.000), (1.375 ± 0.125), and (1.125 ± 0.125) h, respectively, AUC0-t were (65.995 ± 12.367), (178.806 ± 33.592), and (422.836 ± 72.344) h∙ng/mL, respectively, MRT were (2.002 ± 0.061), (1.911 ± 0.042), and (1.955 ± 0.053) h, respectively; The main pharmacokinetics parameters of hyperlipidemia rats after ig administration with low-, medium- and high-dose Sal B were as follows: Cmax were (49.265 ± 7.317), (113.986 ± 15.294), and (246.454 ± 30.476) ng/mL, tmax were (1.125 ± 0.000), (1.125 ± 0.125), and (1.375 ± 0.125) h, respectively, AUC0-t were (96.013 ± 15.384), (207.192 ± 32.676), and (486.843 ± 89.276) h∙ng/mL, respectively, MRT were (2.161 ± 0.049), (2.089 ± 0.033), and (2.097 ± 0.035) h, respectively. Conclusion: The LC-MS method is suitable for the analysis with the high sensitivity and strong specificity. The pharmacokinetics parameters of Sal B of hyperlipidemia rats, such as Cmax, AUC0-t, and MRT, are higher than those of normal rats, which is conducive to play a therapeutic role, but without statistical significance.

ABSTRACT

Objective: To carry out the pharmacokinetic study in awake animal after multiple dosing of Danshen (Salvia miltiorrhiza) Decoction by blood microdialysis method combined with LC-MS. Methods: The pharmacokinetic study was carried out with awake rabbits as experimental animal, Danshen Decoction as tool drug, and Danshensu as evaluation index combining with microdialysis sampling and LC-MS detection technology after intraday multiple dosing. The pharmacokinetic parameters were obtained by non-compartmental model method. Results: The pharmacokinetic study pattern of multiple dosing was established in awake animal, and the whole process dynamic characteristics were collected. The method was stable, reliable, and simple. Conclusion: This study can make the reference to similar study for other drugs.

ABSTRACT

Objective To investigate the iodine nutritional level and thyroid function of pregnant and lactating women in rural areas of Jilin province. Methods The investigation sites were selected from rural areas of three towns (Baoshan, Mingcheng and Yantongshan of Panshi county, Jilin province) in 2009. The pregnant and lactating women were selected as subjects in these three towns. The blood samples were collected and the thyroid function (including serum TT3, TT4, FT3, FT4) were measured with chemiluminescence, and serum thyroglobulin antibodies(TgAb), thyromicrosome antibody(TMAb), and thyroglobulin(Tg) were measured with radioimmunoassay (RIA). The urine samples were collected three times within one month and were measured for iodine concentration by As-Ce catalytic spectrophotometry method. Results In the pregnant women, serum TT3 was higher than that of healthy pregnant women, accounted for 14.3%(8/56), while serum TT4, TT3, FT4 were lower than those of healthy pregnant women, accounted for 3.6%(2/56),5.4% (3/56), and 1.8%(1/56), respectively. In the lactating women,serum TT3 was higher than that of healthy lactating women, accounted for 3.6%(2/56), while serum TT4, FT4 were lower than those of healthy lactating women, accounted for 1.8%(1/56), respectively. Five per cent to 20% of the pregnant and lactating women had higher TgAb and TMAb. Conclusions Existing salt iodine level is appropriate for pregnant women and lactating women, but there was a tendency towards hypothyroid in some women. Routine monitoring of urinary iodine and thyroid function should be carried out among pregnant and lactating women.

ABSTRACT

<p><b>AIM</b>To study the effects of oxyphenamone (Oxy) on activation of Ca(2+)-activated K+ channels in rabbit mesenteric vascular smooth muscle cells.</p><p><b>METHODS</b>To measure the effect of Oxy on the Ca(2+)-activated K+ channel (BK (Ca) channel) activity in rabbit mesenteric vascular smooth muscle cells by using whole cell patch clamp techniques.</p><p><b>RESULTS</b>Oxy reversibly increase BK (Ca) channel activity in rabbit mesenteric artery smooth muscle cells. Application of Oxy (0.1 mumol.L-1) to the perfusion solution caused significant increase in outward currents and its effect was completely abolished by washout; The outward currents K+ was inhibited by TEA (7.5 mmol.L-1); Oxy activated the BK (Ca) channel in a dose-dependent manner (0.01-10 mumol.L-1).</p><p><b>CONCLUSION</b>Oxy directly increase the activity of BK (Ca) channel activity in rabbit mesenteric vascular smooth muscle cells in dose-dependent manner.</p>
